Synapse formation of the AIY neuron on the nerve ring system is studied here, which happens in \"zone 2\" where synaptic vesicle marker Rab3 selectively accumulates (Fig1B and C). Upon disruption of the MIG-10 gene (RAPH1 in humans, protein is known as Lamellipodin), Rab3 localisation in zone 2 is fragmented, suggesting that synaptic vesicles are not at the right location (Fig2 C and D), this can be rescued by expressing the splice variant MIG-10B, MIG-10A and -10C can not do this (Fig2E-J, quantified in Fig2B). Active zone components SYD-1 and SYD-2 show a normal distribution to zone 2 upon MIG-10 deletion (but not quantified; support reference PMID 22451697 Fig6), suggesting that the synapse is formed but vesicles are not clustered to this region. Fig5 shows that, when these active zone components are deleted this also results in MIG-10 dislocation, suggesting that formation of the active zone happens upstream of MIG-10 dependent synaptic vesicle clustering.\n\n8/7/2017 Pim\n- Accession was changed to P34400-2 based on the splice B-variant that specifically mediates the effect on presynapse development..
